泛昔洛韦应用领域

用途一:抗病毒药,用于治疗带状疱疹和生殖器疱

泛昔洛韦制备方法

方法1:0.5mol 2-氨基-6-氯嘌呤溶于含50g氢氧化钠的500ml水中,在10g钯-炭存在下,于0.7MPa氢压和50℃氢化3h,得到83%收率的2-氨基嘌呤。2-氨基嘌呤、2-(2-碘乙基)丙二醇二乙酯和碳酸钾在二甲基甲酰胺中,在室温下搅拌18h,得58%收率的泛昔洛韦。方法2:2-(2-碘乙基)丙二醇二乙酸酯、2-氨基-6-碘嘌呤和碳酸钾在二甲基甲酰胺中,搅拌18h,得到79.4%收率的化合物(I),再在钯-炭催化下,于乙醇中氢化还原得到泛昔洛韦。
方法3:22.5g鸟嘌呤、88g三乙基甲基氯化铵和氯化亚砜,在50~70℃下缓慢搅拌0.5h,再于70℃下保持0.5h,得到28.5g 8-氯鸟嘌呤,其中含14.1%的水。将8-氯鸟嘌呤转化为盐酸盐,并在五氯化二磷存在下干燥。往该盐酸盐(2.04g)中加入6.6g三乙基甲基氯化铵在1lml乙腈的溶液,加入5.6ml三氯氧磷,在60℃下加热1h,得到1.78g 2-氨基-6,8-二氯嘌呤。5.8g 2-氨基-6,8-二氯嘌呤、9.4g 2-(2-碘乙基)丙二醇二乙酸酯和5.9g碳酸钾在100ml二甲基甲酰胺中,在室温下搅拌过夜,得到4.7g化合物(Ⅱ)。3.9g化合物(Ⅱ)在5%钯-炭存在下,以0.35MPa的氢压氢化还原,得到2.4g泛昔洛韦。
